Transplantation of mesenchymal stem cells to treat pronounced smooth tissue defects in congenital malformations.

The fetus is a source of stem cells, with potential applications in medicine. Cells derived from the fetus have more potential than stem cells derived from adult tissues.
Structural defects can be diagnosed with prenatal imaging, and at the time of invasive sampling, like amniocentesis, there is an opportunity to obtain autologous cells from the fetus without any additional risk to the mother or fetus. Mesenchymal stem cells can then be isolated, propagated and characterized during the remainder of the pregnancy, and later utilized for tissue engineering of graft material that will be used for postnatal reconstruction.

Fetal mesenchymal stem cells from amniotic fluid, muscle, skin and placenta will be analysed and compared to each other to see which tissue is superior for tissue engineering. The characterisation consists of investigations of growth kinetics, population potential, colony forming unit-fibroblast, phenotype, immunological properties and in vitro and in vivo differentiation and migration potential. Graft composition and survival of the cells in the graft in vitro and in vivo will be thoroughly examined. Safety of the cell transplantation will be particularly examined using karyotyping, comparative genomic hybridization and tumor formation in vivo.

This whole task is a PhD project, not all of the work described above will be included in the exam thesis. I prefer if the person that takes on this task is willing to do a PhD on the same project.
