The role of non-coding regulatory RNA in the development of Dictyostelium discoideum

The aim of this project is to identify and characterize regulatory non-coding RNA in Dictyostelium discoideum. Recently research concerning small non-coding RNA molecules has acquired increasing interest. These RNA molecules, e.g. snoRNA, snRNA, and micRNA are involved in a variety of different processes such as processing, methylation, and nucleotide modification of rRNA as well as post-transcriptional regulation of mRNA. They function by binding to their target RNA via antisense interaction. It has been suggested that the absence of certain snoRNAs, expressed in the brain, is involved in a severe human disease. We have recently isolated several small RNA, with unknown functions from Dictyostelium discoideum - a social amoeba that goes through a defined developmental program. The project involves expression studies of the isolated RNAs by Northern blot techniques, characterizing precursors by different methods, e.g. 5RACE, and bioinformatic approaches to identify target RNAs upon which these small RNA exert their function. This project is part of a collaboration with Uppsala and Stockholm Universities.
