Genome-wide analysis of developmentally controlled gene expression in a differentiating prokaryote

We are using DNA microarrays to analyse gene expression during cell differentiation in the Gram-positive bacterium Streptomyces coelicolor. Streptomycetes are of immense biotechnological and industrial importance as producers of a wide range of antibiotics and other valuable compounds. In addition, they go through pronounced cell differentiation, leading to both antibiotic production and formation of spores. S. coelicolor is an excellent model organism for studying molecular mechanisms involved in differentiation and determination of cell fate. The genome sequence of S. coelicolor is available, enabling the use of large-scale analysis of the expression-patterns of all its genes during cell differentiation. We will use DNA microarrays to analyse the global patterns of gene expression at
different stages of development. In particular, genes will be identified that are induced during sporulation and that are dependent on any of the hitherto identified regulatory genes that control the process. This project is done in collaboration with Dr. M. J. Buttner, John Innes Centre, Norwich, UK. We are specifically searching for the genes directly involved in modifying or controlling growth, cell cycle progression and morphogenesis during development, and will identify those using genetic approaches among the genes found to be expressed early during sporulation.
